Description

Usually histologic sections should have a uniform thickness. Sometimes thickness may alternate between too thick and too thin.


 

Possible Explanation

Solution

The paraffin is too soft.

Consider re-embedding in a harder paraffin. Remove any excess paraffin from the blade after each advance.

The blade is loose in its holder.

Tighten the blade in its holder.

The block is loose in its holder.

Tighten the block in its holder.

There is a problem with the microtome advance mechanism.

Keep the microtome in good working order. Examine for worn or broken parts.

The clearance angle is too small.

Increase the clearance angle.
