Two immunostains may be performed simultaneously for a number of reasons. The reasons determine how to quantitate the cell population(s).
Parameters:
(1) percent of cells staining with X
(2) percent of cells staining with Y
Situation 1: One population marked by Y. There is another population of cells which has a subpopulation marked by X.
non-Y population = 100 - Y
percent of non-Y population marking with X =
= X / (100 - Y) * 100%
Situation 2: One population marked by Y, with a subpopulation of this group marked by X
non-Y population = 100 - Y
percent of Y population marked with X =
= X / Y * 100%
Example: Bone marrow biopsy involved with malignant lymphoma, with one marker for malignant lymphoma and the second marker for a residual marrow element.
