Description

After a bolus (rapid) intravenous infusion of drug, the plasma concentration usually reaches a maximum concentration quickly, after which it declines. For many drugs this occurs as a first order process. The primary differences from oral dosing are that there is no absorption phase and the total dose is delivered.


 

Parameters:

(1) dose in mg

(2) volume of distribution in liters

(3) plasma concentration in mg/L

(4) peak plasma concentration just after the infusion in mg/L

(5) elimination rate constant per hour

(6) time since the bolus dose in hours

 

peak plasma concentration in mg/L =

= (dose administered) / (volume of distribution)

 

plasma concentration at a given time after the bolus dose =

= (peak plasma concentration in mg/L) * EXP((-1) * (elimination rate constant per hour) * (time in hours)
