The PILE score is predictive of prognosis for a cancer patient treated with immune checkpoint inhibitors anti-PD-1/anti-PD-L1.
PILE is acronym for PIV, LDH and ECOG.
Parameters prior to administration of first dose of immune checkpoint inhibitor):
(1) blood PIV (Pan-Immune-Inflammation Value)
(2) median value for blood PIV
(3) serum LDH in U/L
(4) upper limit of normal (ULN) for serum LDH
(5) ECOG status

serum LDH
<= ULN

ECOG status

PILE score =
= SUM(points for all 3 parameters)
Interpretation:
• minimum score: 0
• maximum score: 3
• A score of 2 or 3 is high risk and is associated with a decreased overall survival.
Performance:
• The area under the ROC curve is 0.65 for overall survival.
