Description

Wong et al developed a simplified method for monitoring the area under the curve for serum tacrolimus in a renal transplant recipient. This allows the monitoring to be done with a minimum number of specimen collections and may be more useful than monitoring the morning trough level. The authors are from Queen Elizabeth Hospital in Hong Kong.


 

Tacrolimus = FK506, Prograf (Fujisawa)

 

Parameters:

(1) whole blood concentration at 2 hours in ng/mL

(2) whole blood concentration at 4 hours in ng/mL

 

predicted 12 hour area under the curve in hour • ng/mL =

= (2.4 * (concentration at 2 hours)) + (5.9 * (concentration at 4 hours)) + 16.2

 

Target value: 100 hr • ng/mL (avoids excessive adverse events while providing adequate immunosuppression according to Filler et al).

 

Performance:

• R-squared: 0.93

• Percentage of prediction error (PE%): -0.2% (range -13% to +9.3%)
