Description

Weber et al developed an abbreviated pharmacokinetic protocol for monitoring serum levels of mycophenolic acid (MPA), which is the active component of the immunosuppressive agent mycophenolate mofetil (MMF). This can provide a reasonable pharmacokinetic profile with a minimum of samples. The authors are from the Children's Hospitals associated with the University of Heidelberg and Ludwig-Maximillians University (Munich), as well as the Georg-August University of Gottingen in Germany.


 

Patient selection: pediatric patient with a renal transplant being treated with MMF

 

Concurrent medications: cyclosporine and prednisone

 

Parameters:

(1) plasma concentration of MPA at T0 (immediately prior to dosing) in mg/L

(2) plasma concentration of MPA at 30 minutes after dosing in mg/L

(3) plasma concentration of MPA at 2 hours after dosing in mg/L

 

area under the curve for the serum concentration of MPA for 0 to 12 hours after dosing in mg/L•h =

= (4.3 * (concentration at T0)) + (0.54 * (concentration at 30 minutes)) + (2.15 * (concentration at 2 hours)) + 18.6

 

Performance:

• The coefficient of determination (r^2) was 0.76.

• The percentage prediction error was 10.7%.
