Description

Proteins in solution absorb ultraviolet light at 280 nm primarily due to the presence of tryptophan, tyrosine and phenylalanine.


Light source: ultraviolet

 

Parameters:

(1) absorption at 280 nm from 0 to 1.0

(2) absorption at 260 nm from 0 to 1.0 (which removes interference from nucleic acids)

 

protein concentration in mg/mL =

= (1.55 * (absorption at 280 nm)) - (0.76 * (absorption at 260 nm))

 

Interpretation:

The applicable protein concentration range is <= 1.5 mg/dL.

The absorbance may be affected by pH, ionic strength and the presence of other chemicals.
