Sometimes it may be difficult to confirm the presence of vas deferens in histologic sections of a vasectomy specimen. Sasaki et al used CD10 and pankeratin immunostains to help identify a vas with suboptimal histologic features.
Reasons why a specimen may be hard to identify as a vas:
(1) small size
(2) poor orientation
(3) crush artifact
(4) thermal artifact
(5) poor fixation
(6) prior freezing
Immunostaining panel:
(1) CD10 (CALLA): stains Wolffian duct structures
(2) pankeratin
Immunostain
Cell Layer
Appearance if Vas Deferens
CD10
apical
strong membrane staining along the luminal surface
CD10
basal
weaker cytoplasmic staining
pankeratin
apical
positive in membranes and cytoplasm
pankeratin
basal
positive in membranes and cytoplasm
