Description

McCarty et al described a histochemical score that could be used to quantitate the intensity of staining shown by an immunohistochemical tumor marker.

For a histochemical marker, each of the following is recorded:

(1) the intensity of staining from 0 to 4+

(2) the percent of the tumor showing each staining intensity

Staining Intensity

Factor

(x + 1)

Value

0 (none)

NA

0 * (percent of tumor cells negative)

1+ (weak)

2

2 * (percent of tumor staining 1+)

2+

3

3 * (percent of tumor staining 2+)

3+

4

4 * (percent of tumor staining 3+)

4+ (strong)

5

5 * (percent of tumor staining 4+)

where:

• The percent staining is a number from 0 to 100.

• The sum of all the percent staining must equal 100.

• The factor for each level is (x + 1), where x indicates the staining intensity (x+)

histochemical score =

= SUM(values for each level of positive staining)

Interpretation

• minimum score: 0

• maximum score: 500

• For each tumor marker the lowest score determining a positive reaction needs to be determined.
