Description

During the second phase of a biphasic system the drug concentration follows first order kinetics. Sometimes the semilogarithmic line of drug concentration vs time is extrapolated back to time 0 to determine a theoretical initial concentration that would have been present if the drug followed a single compartment model.

= LN(hypothetical concentration of drug at time 0) - ((elimination rate constant) * (T in hours))

= (concentration at time T) * EXP((elimination rate constant) * (time T))
