Description

Aronson and Ferner reported the DoTS classification scheme for adverse drug reactions. The authors are from Radcliffe Infirmary and City Hospital Birmingham.

Parameters:

(1) dose dependency (Do)

(2) timing dependence (T)

(3) susceptibility (S)

Dosing dependence:

(1) overdose (supratherapeutic toxic effects)

(2) standard therapeutic dose (collateral effect)

(3) subtherapeutic dose (hypersusceptibility)

Timing dependent reactions:

(1) reaction to rapid administration

(2) first dose reaction

(3) early reaction (then abate)

(4) intermediate reaction (some delay)

(5) late reaction

(6) delayed reaction

Susceptibility:

(1) genetic factor

(2) age

(3) sex

(4) altered physiology

(5) exogenous factor (includes drug iinteraction)

(6) disease (renal or other organ failure)
