Description

Restriction enzymes will digest double stranded DNA into smaller fragments. The moles of the fragment ends can be calculated from the amount of DNA present and the number of cuts.


 

Parameters:

(1) moles of double stranded DNA

(2) number of cuts made

(3) linear vs circular DNA

 

molecular mass of a double stranded DNA fragment =

= (649 grams per mole • base pair) * (number of base pairs)

 

moles of ends of linear DNA =

= 2 * (grams of DNA) / (molecular mass of the double stranded DNA fragment)

 

where:

• 2 indicates the number of ends in a linear stand of DNA.

 

For circular DNA:

 

moles of ends =

= (2 * moles of DNA) * (number of cuts)

 

For linear DNA

 

moles of ends =

= (2 * moles of DNA) * ((number of cuts) + 1)
