The dead volume of a chromatographic column can be calculated from the column dimensions. If the flow rate is known, then the dead time can be estimated.
Parameters:
(1) length of the column in cm
(2) inner diameter of the column in cm
(3) flow rate through the column
For the column:
dead volume in mL =
= 0.5 * ((diameter in cm)^2) * (column length in cm)
If the flow rate is known, then:
dead time in minutes =
= (dead volume in mL) / (flow rate in mL/min)
The retention factor for a compound can be calculated as:
retention factor =
= ((retention time for the compound) - (dead time)) / (dead time)
If the retention factor is between 1 and 20 then the chromotographic system should give acceptable performance.
