Description

A section stained with hematoxylin & eosin may show blue staining of tissues that are expected to be eosinophilic.


Stain: hematoxylin and eosin

 

Situation: bluish staining of collagen, smooth muscle and collagen

 

Troubleshooting:

(1) The hematoxylin solution has a pH > 3.0.

(2) The hematoxylin formulation is not selective enough.

(3) There was inadequate differentiation.

(4) The acid-alcohol is too weak.

(5) The time spent in the acid-alcohol was too short.

(6) The eosin stain is contaminated with the bluing solution.

(7) The slides spent too much time in the dehydrating or differentiating alcohols (with excessive removal of the eosin).
