Nucleic acids in solution can be quantitated using absorbance by ultraviolet (UV) spectrophotometry.


 

Testing instrumentation:

(1) ultraviolet spectrophotometer

(2) cuvettes with 1 cm light path

Concentration Giving an Absorbance of 1.0 at 260 nm

double stranded DNA

50 µg/mL

single stranded DNA

33 µg/mL

single stranded RNA

40 µg/mL

 

concentration of nucleic acid in µg/mL =

= (concentration giving absorbance at 1.0) * (absorbance of solution at 260 nm)
